The practice of microdosing psychedelics, specifically LSD, has garnered attention due to its purported therapeutic effects. The researchers conducted a systematic review of 14 relevant studies focusing on laboratory-controlled trials of microdose LSD, defined as doses between 5-20 micrograms. The aim of the review was to outline recent experimental findings while identifying gaps in the existing literature and limitations in methodology to inform future research efforts.

Study Demographics and Methodology

The trials primarily involved young, healthy men and women aged 22-38, with a balanced representation of genders. However, studies did not consider variations related to gender or menstrual cycles. All participants received either LSD or a placebo, while doses generally ranged from 5g to 20g. Some research included a titration approach, allowing participants to adjust to a comfortable dose.

Physiological and Psychological Effects

Data from two studies illustrated the pharmacokinetics of LSD, noting how plasma concentration peaked comparably across doses and timeframes. Most trials reported mild adverse effects, with headaches and anxiety as common reactions, leading to withdrawal in a small percentage of cases. Physiologically, low doses of LSD generally elevated systolic blood pressure and introduced various neural connectivity changes. Effects included enhanced connectivity in brain regions related to emotion and cognition. Behavioral assessments indicated increased positivity, decreased negative responses in social rejection scenarios, and improved pain tolerance. Notably, cognitive performance and creativity did not seem significantly affected by low doses, although subjective reports reflected enhanced feelings of connection and emotional positivity.

Changes with Repeated Dosing

The research indicated that although initial effects were more prominent, repeated microdosing did not yield improvements in overall mood or cognitive abilities. The researchers emphasized that individuals reacted differently to LSD dosages due to various factors, including pre-existing mood states and genetic differences in drug response. There is a need for attention to these individual variances when evaluating microdosing effects.

Future Research Directions

Despite generating mild acute effects on mood and cognition, the evidence does not support longer-term psychological benefits of microdosing LSD. The findings highlight the necessity for additional clinical trials to explore its therapeutic applications thoroughly, particularly in treating mood disorders. The current lack of evidence showing substantial cognitive enhancements calls for more rigorous assessments of claims around creativity and improved cognitive functions related to microdosing. Future studies should aim to clarify the impact of microdoses on individuals with varying baseline characteristics and understand the underlying neurobiological mechanisms involved.
